Live Cell Tomography

Optical tomography of single living cells, cell clusters and cell spheroids is reported. The latter ones often serve as tumour models or as test systems for pharmacological agents. However, even upon multiple exposures, light dose has to be limited to avoid phototoxicity or cell damage. In view of these restrictions we developed the following experimental methods:
- Light Sheet based Fluorescence Microscopy (LSFM);
- Axial tomography for 3D imaging from various sides;
- Structured Illumination Microscopy (SIM) for obtaining ultimate resolution;
- Variable-angle Total Internal Fluorescence Microscopy (VA-TIRFM) for cell membrane topology.
